When inflammation fails to resolve, tissue repair stalls

Inflammation is an essential part of the normal healing response. Following injury, immune cells coordinate pathogen defence, debris clearance and early tissue repair. However, successful healing depends on the timely transition from inflammation to resolution.

In chronic non-healing wounds, this transition fails. The wound environment remains dominated by persistent inflammatory signalling, immune cell dysfunction, impaired angiogenesis and defective extracellular matrix remodelling. As a result, the tissue becomes trapped in a cycle of inflammation, damage and failed regeneration.

This failure of resolution is a central biological bottleneck in chronic wound healing and is increasingly recognised as a broader driver of chronic inflammatory disease.

Restoring the body’s pro-resolution pathways

Many current anti-inflammatory treatments focus on suppressing inflammatory pathways. While this can reduce inflammatory signalling, it does not necessarily restore the active biological programmes required for resolution and repair.

EVolution Therapeutics is taking a different approach. Our platform is designed to deliver mRNA instructions for pro-resolving proteins directly into diseased tissue, enabling local cells to produce therapeutic signals that help shift the tissue environment from chronic inflammation towards healing.

By supporting endogenous pro-resolution biology, our aim is to promote a coordinated repair response rather than simply blocking inflammation.

Local delivery designed for sustained tissue exposure

Our therapeutic platform combines mRNA-loaded lipid nanoparticles with a biodegradable hydrogel formulation for local delivery at the site of disease.

The lipid nanoparticles are designed to protect and deliver therapeutic mRNA into local target cells. The hydrogel acts as a local depot, supporting retention of the formulation within the wound environment and enabling sustained exposure to therapeutic nanoparticles over time.

This approach is designed to concentrate biological activity where it is needed most, while reducing reliance on systemic immune modulation.

Chronic non-healing wounds as our first clinical focus

Chronic non-healing wounds provide a clinically important and biologically relevant first indication for our platform. These wounds are characterised by persistent inflammation, impaired vascularisation, disrupted tissue architecture and failure to progress through the normal phases of healing.

Current care often focuses on managing the wound environment through dressings, debridement, infection control and off-loading. EVolution Therapeutics is developing an active therapeutic approach designed to intervene at a key biological bottleneck: the failure to resolve inflammation and restart repair.

Our preclinical studies support the potential of this approach, with evidence of accelerated wound closure and enhanced tissue repair across wound-healing models.
